Yep, it has to do with stoner culture. I don't know if it refers to the date 4/20 or the time of day 4:20, but it indicates a lot of smokin'.

I'm not sure where 420 comes from, but it indicates both a date and a time. 4:20 is smokeup time for most stoners, which is odd since 4:20 in the morning is too early, and 4:20 in the afternoon is after school. The date 4/20 has more significance than just a stoner day. It was also Hitler's birthday, and the day of the Columbine school shootings.
